Effect of concentration and exposure period to butyrolactone I on meiosis progression in bovine oocytes Arq. Bras. Med. Vet. Zootec.
Adona,P.R.; Leal,C.L.V..
The effect of concentration and exposure period of bovine oocytes to butyrolactone I (BLI) on meiotic block and in vitro maturation (IVM) kinetics was studied. In experiment 1, all oocytes were at germinal vesicle stage (GV), after 6h in culture with 0, 50 and 100µM BLI. After 12h, all oocytes cultured with 50 and 100µM BLI remained in GV. After 24h, less oocytes were in GV with 50µM (82%) than with 100µM BLI (99%, P<0.05). In experiment 2, after 6h IVM, 93% of control oocytes (IVM only) were in GV, while treated oocytes (100µM BLI for 6, 12 or 24h prior to IVM) showed less oocytes in GV with increased exposure period to BLI prior to IVM (83 and 73%, for 6h and 12h, P<0.05). For a 24h inhibition, GV rates were similar to 12h (70%, P>0.05). After...

Gonadal development and reproductive period of the characin Astyanax aff. bimaculatus (Characiformes: Characidae) in a tropical reservoir in southeastern Brazil Rev. Bras. Zool.
Araújo,Francisco G.; Nascimento,Aparecida A.; Gomes,Iracema D.; Sales,Armando; Oliveira,Beatriz A. Chagas de.
ABSTRACT The gonadal development of the characin Astyanax aff. bimaculatus from Funil Reservoir, an impoundment in the middle reaches of the Paraiba do Sul River in southeastern Brazil was analysed through histological, histochemical and histometric techniques, and the spawning season was determined. Unbalanced sex ratio was found, with females outnumbering and reaching larger sizes than males. The following stages of oogenic cell development were identified for females: primary growth, pre-vitellogenic (cortical alveolar) and vitellogenic. In males, the following stages were determined: spermatogonia primary and secondary, spermatocyte primary and secondary, spermatid and spermatozoa. The spawning occurs most of the year, peaking in January-February, as...

Oocyte development in cattle: physiological and genetic aspects R. Bras. Zootec.
Britt,Jack H..
Oocytes in cattle are formed during embryogenesis and develop within individual follicles in the cortex of the ovary. Dormant primordial follicles become active and undergo progressive development at regular intervals commencing during the late fetal stage and continuing throughout adulthood. Once activated, follicles and oocytes in a cohort either grow to maturation and ovulation or undergo atresia, ultimately depleting the ovaries of germ cells. It takes an estimated 100 days for a follicle and its oocyte to reach the mature ovulatory stage. Within an individual, number of follicles in one ovary is similar to number in the other ovary; however, there are large differences among individuals in total number of follicles present in both ovaries. In...

Pre-incubation of porcine semen reduces the incidence of polyspermy on embryos derived from low quality oocytes Ciência Rural
Brogni,Cláudio Francisco; Ohlweiler,Lain Uriel; Klein,Norton; Mezzalira,Joana Claudia; Cristani,Jose; Mezzalira,Alceu.
ABSTRACT: The main cause of low efficiency of in vitro produced porcine embryos is the high polyspermic penetration rates at fertilization, which is aggravated in low quality oocytes. Experiment 1 evaluated the embryo development in high and low quality oocytes. Experiment 2 evaluated the embryo development and quality of low quality oocytes fertilized with sperm pre-incubated during 0h (control), 0.5h, 1h and 1.5h. Experiment 3 investigated fertilization and monospermic rates of the same groups of Experiment 2. Experiment 4 evaluated embryo development, cell density, fertilization and monospermic rates of high quality oocytes using semen pre incubated during the best time observed in the previous experiments. Cleavage and blastocyst rates were analyzed by...

Maternal transmission of mitochondrial diseases Genet. Mol. Biol.
Chiaratti,Marcos R.; Macabelli,Carolina H.; Augusto Neto,José Djaci; Grejo,Mateus Priolo; Pandey,Anand Kumar; Perecin,Felipe; Collado,Maite del.
Abstract Given the major role of the mitochondrion in cellular homeostasis, dysfunctions of this organelle may lead to several common diseases in humans. Among these, maternal diseases linked to mitochondrial DNA (mtDNA) mutations are of special interest due to the unclear pattern of mitochondrial inheritance. Multiple copies of mtDNA are present in a cell, each encoding for 37 genes essential for mitochondrial function. In cases of mtDNA mutations, mitochondrial malfunctioning relies on mutation load, as mutant and wild-type molecules may co-exist within the cell. Since the mutation load associated with disease manifestation varies for different mutations and tissues, it is hard to predict the progeny phenotype based on mutation load in the progenitor. In...

Infection of the cockle Cerastoderma edule in the baie des veys (France) by the microsporidian parasite Steinhausia sp. ArchiMer
Comtet, T; Garcia, Celine; Le Coguic, Yvette; Joly, Jean-pierre.
We report the occurrence of the microsporidian parasite Steinhausia sp. in the oocytes of the common cockle Cerastoderma edule in a natural population in France, where high mortalities occurred. Steinhausia sp. appeared primarily as sporocysts containing many small spores, and putative earlier developmental stages were also observed. Both its prevalence and infection intensity were low, and no host defence reaction was recognized, suggesting that Steinhausia sp. had no detrimental effect on C. edule. Its prevalence was higher in cockles lying on the sediment surface, but the significance of this observation could not be explained given the poor knowledge of the Steinhausia life cycle. The present data did not allow specific identification of the parasite,...

Reactive oxygen species in bovine embryo in vitro production Biocell
Dalvit,G.C.; Cetica,P.D.; Pintos,L.N.; Beconi,M.T..
Oxidative modifications of cell components due to the action of reactive oxygen species (ROS) is one of the most potentially damaging processes for proper cell function. However, in the last few years it has been observed that ROS participate in physiological processes. The aim of this work was to determine ROS generation during in vitro production of bovine embryos. Cumulus-oocyte complexes were recovered by aspiration of antral follicles from ovaries obtained from slaughtered cows and cultured in medium 199 for 22 h at 39ºC in 5% CO2: 95% humidified air. In vitro fertilization was carried out in IVF-mSOF with frozenthawed semen in the same culture conditions and embryo in vitro culture in IVC-mSOF at 90% N2: 5% CO2: 5% O2. ROS was determined in denuded...

A microarray-based analysis of oocyte quality in the European clam Ruditapes decussatus ArchiMer
De Sousa, Joana Teixeira; Milan, Massimo; Pauletto, Marianna; Bargelloni, Luca; Joaquim, Sandra; Matias, Domitilia; Matias, Ana Margarete; Quillien, Virgile; Leitao, Alexandra; Huvet, Arnaud.
A microarray-based analysis was performed with the objective of describing genomic features of oocytes and looking for potential markers of oocyte quality in the economically important European clam, Ruditapes decussatus. Oocytes of 25 females from Ria de Aveiro, Western coast of Portugal (40°42′N; 08°40′W) were selected for this study and oocyte quality was estimated by success of D-larval rate under controlled conditions, which appeared to vary from 0 to 95 %. By genome-wide expression profiling with a DNA microarray, 526 probes appeared differentially expressed between two groups representing the largest and smallest value of D larval rates, named good (represented by a mean D-larval yield of 57 ± 22%) and poor (9 ± 5%) quality oocytes. Enrichment...

The nme gene family in zebrafish oogenesis and early development ArchiMer
Desvignes, T.; Fauvel, Christian; Bobe, J..
After the recent report of the expression of several nme genes in the zebrafish gonads, the present study aimed at further analyzing the expression of nme genes in the ovary with special attention for the nme transcripts that are maternally inherited and could thus participate in the determination of oocyte developmental competence. The expression levels of all groups I and II nme genes were characterized by QPCR in a panel of zebrafish tissues. The nme genes exhibiting an ovarian expression were subsequently monitored throughout oogenesis and early development, and their expression sites characterized using in situ hybridization. Here, we show that nme2b1, nme3, nme4, and nme6 are highly expressed in the ovary and present in the zebrafish oocyte...

The Nme gene family in fish ArchiMer
Desvignes, T.; Fostier, A.; Fauvel, Christian; Bobe, J..
The Nme gene family, also known as Nm23 or NDPK, is a very ancient gene family that can be found in all kingdoms of life. In the late eighties, a gene of the Nme family, NME1, was identified as the first metastatic suppressor gene, resulting in a major interest for this family. Due to the complexity of the family, the need for a unified and evolutionary-supported gene nomenclature was recently stressed by the scientific community. Based on a complete evolutionary history study of the gene family in metazoans and vertebrates, a unified nomenclature was recently proposed and accepted by gene nomenclature consortia. In addition to its well-documented role in tumor metastasis, members of the Nme family are also involved in a wide variety of cellular and...

X-Linked Retinitis Pigmentosa 2 Is a Novel Maternal-Effect Gene Required for Left-Right Asymmetry in Zebrafish ArchiMer
Desvignes, Thomas; Nguyen, Thaovi; Chesnel, Franck; Bouleau, Aurelien; Fauvel, Christian; Bobe, Julien.
Retinitis pigmentosa 2 (RP2) gene is responsible for up to 20% of X-linked retinitis pigmentosa, a severe heterogeneous genetic disorder resulting in progressive retinal degeneration in humans. In vertebrates, several bodies of evidence have clearly established the role of Rp2 protein in cilia genesis and/or function. Unexpectedly, some observations in zebrafish have suggested the oocyte-predominant expression of the rp2 gene, a typical feature of maternal-effect genes. In the present study, we investigate the maternal inheritance of rp2 gene products in zebrafish eggs in order to address whether rp2 could be a novel maternal-effect gene required for normal development. Although both rp2 mRNA and corresponding protein are expressed during oogenesis, rp2...

Spawning energetics and otolith microchemistry provide insights into the stock structure of bonga shad Ethmalosa fimbriata ArchiMer
Doering, Julian; Wagner, Carola; Tiedemann, Maik; Brehmer, Patrice; Ekau, Werner.
The gross energy content of spawning batches and the microchemistry of sagittal otoliths in individual female bonga shad Ethmalosa fimbriata were compared between contrasting sampling sites at the Senegalese southern coast and inside the hypersaline Sine Saloum Estuary. Results show that females spawning in the estuary's middle reaches invested almost three times more energy into reproduction (115 +/- 65 J g(-1) body mass) than their neritic counterparts (39 +/- 34 J g(-1) body mass). Also, female otolith levels of Ba:Ca, Sr:Ca and Zn:Ca either differed significantly between study sites or could be linked to heterogeneous environmental variables. A quadratic discriminant function analysis provided evidence of segregated spawning populations of E. fimbriata...

In vitro fertilization of the rock shrimp, Rhynchocinetes typus (Decapoda, Caridea): a review Biol. Res.
Dupré,Enrique M; Barros,Claudio.
This review compiles all the research done on gametes and fertilization in the rock shrimp, R. typus, and describes the sequence of events from the first gamete interaction to zygote formation and the first cleavage of the embryo, with light, fluorescence confocal and electron microscopes. Early studies showed that sperm from the vas deferens have a tack-shape with a "needle-like process" or rigid spike (RS) that extends from a semi-spherical body that contains the arms with chromatin and spines. Upon contact with seawater and by action of Na +, the arms and spines extend, producing an inverted umbrella form of the spermatozoa. The first sperm-oocyte interaction occurs between protein receptors type lectins of the sperm RS and oocyte chorion sperm ligands....

Transfection of Gametes: A Method to Generate Transgenic Animals International Journal of Morphology
Esponda,Pedro.
The production of transgenic animals (TA) using transfected spermatozoa or eggs is commented. Different methods have been employed to introduce transgenes into the gametes of several vertebrates and invertebrates. Methods for the transfection of gametes have employed naked DNA, viral vectors, DNA/Liposome complexes, electroporation or high velocity microprojectiles. Spermatozoa and oocytes or eggs have showed a good transfection efficience (80% in some cases), and microscopical observations demonstrated that the transgenes appeared localized in the nucleus. Gametes have shown to be naturally protected against the entrance of foreign genes because some semino plasma or plasma membrane proteins block the entrance of foreign genes in spermatozoa. In the...

Morphological and Morphometric Aspects of Crassostrea rhizophorae (Guilding, 1828) Oocytes in Three Stages of the Gonadal Cycle International Journal of Morphology
Ferreira,M. A. P.; Paixão,L. F.; Alcântara-Neto,C. P.; Santos,S. S. D.; Rocha,R. M..
Crassostrea rhizophorae (Guilding, 1828) is one of the tropical species of oysters cultivated on the Brazilian shore. Despite its high commercial value, there is no information on the gonadal characteristics during the reproductive stages of this species. The objective of this work was to analyze the morphology and morphometry of Crassostrea rhizophorae oocytes and follicles in three stages of the gonadal cycle. Were analized histological sections of gonads considering that the oocytes presented visible nuclei and nucleoli, it was observed three gonadal cycle stages: early gametogenesis, growth and maturation. In the early gametogenesis stage, follicles and oocytes presented diameters of 180.29 (± 41.91) and 18.66 (± 6.85) µm, respectively, and presence of...

Ovary Development and Maturation in Nezara viridula (L.) (Hemiptera: Pentatomidae) Neotropical Entomology
Fortes,P; Salvador,G; Cônsoli,FL.
Ovary development and maturation of Nezara viridula (L.) were evaluated by examining ovariole morphology and the alterations in the biochemical (protein synthesis related to reproduction) composition of the hemolymph. Quantitative and qualitative protein analyses were performed and ovary structural alterations for the pre-reproductive and reproductive stages were recorded. Total concentration of proteins in female hemolymph gradually increased until the end of the pre-mating stage, remaining unaltered thereafter. Proteins linked to reproduction (vitellogenins) appeared in the hemolymph 10 days after adult emergence and indicated the end of the pre-mating stage. After mating, total protein concentration in the hemolymph was lower compared to virgin females;...
